5-AF594 tyramide 

5-AF594 tyramide is a red fluorescent dye (Ex=590 nm, Em=618 nm). 5-AF594 tyramide is utilized as reporter fluorescent substrate of horseradish peroxidase (HRP)-catalyzed deposition for tyramide signal amplification (TSA). 5-AF594 tyramide can be used for multiplex immunohistochemistry (mIHC)^[1].

5-AF594 tyramide 检测
1) 切片在室温下用 0.25% Triton X-100 通透 10 分钟。
2) 一抗 4°C 孵育过夜。
3) 在 3% H₂O₂ 的1x PBS 中的孵育 20 分钟，以阻断内源性过氧化物酶活性。
4) 用 FITC 偶联的抗兔 IgG 一抗在室温下 (1:100) 孵育1 小时，然后用 FITC 偶联的 HRP 抗体 (1:500) 在 4°C 下孵育过夜。
5) 用 5-AF594 tyramide 孵育 10 分钟 (1:200)。
6) 荧光显微镜检测。
注：建议根据实际情况确定 5-AF594 tyramide 工作液的最佳浓度。
