5-FITC tyramide 

5-FITC tyramide is a green fluorescent dye composed of FITC (HY-66019) and tyramide (Ex=488 nm, Em=525 nm). 5-FITC tyramide is utilized as reporter fluorescent substrate of horseradish peroxidase (HRP)-catalyzed deposition for tyramide signal amplification (TSA). 5-FITC tyramide can be used for multiplex immunohistochemistry (mIHC)^[1].

5-FITC tyramide 检测
1) 将 4 μL WGA-HRP 示踪剂注射至大鼠神经节，然后获取特定组织用于切片。
2) 将切片干燥 60 分钟。
3) 在 TNT 缓冲液 (0.1 M Tris-HCl、0.15 M NaCl、0.05% Tween 20，pH 7.5) 中复水，然后在新鲜配制的 NaBH4 (0.5 mg/mL) 的 TN 缓冲液 (0.1 M Tris-HCl、0.15 M NaCl，pH 7.5) 中淬灭 15 分钟。
4) 用 TNT 清洗，然后用 0.1% Triton X-100 的 TN 缓冲液通透 15 分钟。
5) 用加 2% BSA 的 TNT (TNT-BSA) 孵育 60 分钟，用 TNT 洗涤后，加入 5-FITC tyramide (扩增稀释液 1:50 稀释) 孵育 15 分钟。
6) TNT 洗涤后，加入一抗 4°C 过夜孵育，加入驴抗兔 IgG 标记的 Lissamine rhodamine (LRSC) 或 Cy3 (HY-D0822) (TNT-BSA 1:200 稀释) 和驴抗羊标记的 Cy5 (HY-D0821) (TNT-BSA 1:200 稀释)。
7) 荧光显微镜检测。
注：建议根据实际情况确定 5-FITC tyramide 工作液的最佳浓度。
