8pyDTZ 

8pyDTZ is a pyridyl diphenylterazine (DTZ) analog and an ATP-independent pyridyl substrate of LumiLuc luciferase. 8pyDTZ exhibits spectrally shifted emission. 8pyDTZ has excellent biocompatibility and superior in vivo sensitivity. 8pyDTZ can be used for in vivo luminescence imaging^[1].

1 工作液制备^[2]
1) 在含 10% Ethanol (v/v)，10% Glycerol (HY-B1659) (v/v)，10% 2-Hydroxypropyl-β-cyclodextrin (HPBCD, w/v) 和 35% PEG-300 (HY-Y0873) (v/v) 的水性可注射制剂中，制备 8pyDTZ 溶液。
2) 典型工作液为 480 μL 制剂中含 4 μmol (1.5 mg) 8pyDTZ。

2 具体染色步骤
1) 经异氟烷吸入麻醉小鼠 5 min，用电剃刀去除成像区域毛发，涂抹 Nair 脱毛膏 1 min 后以无菌纱布擦净残留毛发，用 DPBS 冲洗并以 70% 乙醇消毒。
2) 将 8pyDTZ 工作液腹腔注射至小鼠左下腹腹腔内。
3) 立即将小鼠置于生物发光成像系统 (如 Ami HT 系统) 中，设置参数：发射滤光片全开，视野 25 cm，光圈 f-stop 1.2，合并模式 1×1，曝光时间 1 s。
4) 每 1 min 采集一次生物发光图像，持续 20 min。使用 Fiji (ImageJ) 软件分析图像，并用 Excel 进行数据处理。
