Eriochrome black T, Indicator is a complexing agent for metal ions (e.g., Ca2+, Mg2+) and is used as an indicator in complexometric titrations. Eriochrome black T, Indicator forms colored complexes with metal ions through covalent coordination bonds, and indicates the endpoint of the titration by color change. Eriochrome black T, Indicator can be used as an anionic azo dye in photocatalytic degradation studies to evaluate the performance of photocatalysts. The reaction solution of Eriochrome black T, Indicator combined with Mg2+ is initially purple. During loop-mediated isothermal amplification (LAMP), the color changes from purple to sky blue due to the consumption of Mg2+ by the formation of magnesium pyrophosphate, indicating a positive reaction. The optimal concentration of Eriochrome black T, Indicator in LAMP is 60 μM, and the detection limit for Mycobacterium tuberculosis is 1 pg DNA/reaction[1][2].

Eriochrome black T, Indicator 可用于环介导等温扩增 (LAMP) 反应检测结核分枝杆菌，通过 LAMP 反应中的镁离子消耗情况，间接指示细菌 DNA 的存在。Eriochrome black T, Indicator 可与 Mg2+ 结合形成紫色络合物。在 LAMP 反应初始阶段，溶液中 Mg2+ 浓度充足，Eriochrome black T, Indicator 呈现紫色。等产生副产物焦磷酸镁 (Mg2P2O7) 沉淀，Eriochrome black T, Indicator 变成天蓝色[2]。

1. DNA 提取
(1) 生物标本处理：
采用 Petroff 法处理痰液或支气管肺泡灌洗液，通过煮沸法提取 DNA。
用 TE 缓冲液洗涤生物标本 2 次，沉淀煮沸 5-10 分钟，离心后取上清作为模板 DNA。
(2) 纯化 DNA 制备：
从结核分枝杆菌 H37Rv 菌株中提取基因组 DNA，用于优化反应和检测限测定：菌液悬浮于 TE 缓冲液，80°C 灭活 1 小时，37°C 用溶菌酶和蛋白酶 K 裂解，经氯仿-异戊醇抽提和异丙醇沉淀获得纯化 DNA。

2. LAMP 反应体系 (以 25 μL 体系为例)
反应组分：
内引物 (FIP、BIP) 各 1.6 μM，外引物 (F3、B3) 各 0.2 μM，环引物 (FLP、BLP) 各 0.8 μM；
0.8 M 甜菜碱，1X ThermoPol 缓冲液，8 U Bst DNA 聚合酶，1 ng 模板 DNA；
3.5 mM MgSO4，1.4 mM dNTPs，60 μM EBT 指示剂。
孵育条件：64°C 恒温孵育 30 分钟，无需变性或退火步骤。

3. 结果观察
阳性判定：反应液颜色从紫色变为天蓝色，表明存在目标 DNA。
阴性对照：不含模板 DNA 的反应液应保持紫色。

    请依序添加每种溶剂： 10% DMSO    40% PEG300    5% Tween-80    45% Saline

    Solubility: ≥ 2 mg/mL (4.33 mM); 澄清溶液

    此方案可获得 ≥ 2 mg/mL（饱和度未知）的澄清溶液。

    以 1 mL 工作液为例，取 100 μL 20.0 mg/mL 的澄清 DMSO 储备液加到 400 μL PEG300 中，混合均匀；再向上述体系中加入 50 μL Tween-80，混合均匀；然后再继续加入 450 μL 生理盐水 定容至 1 mL。

    生理盐水的配制：将 0.9 g 氯化钠，溶解于 ddH₂O 并定容至 100 mL，可以得到澄清透明的生理盐水溶液。
