p-Phenetidine (4-Ethoxyaniline) is a metabolite that participates in the oxidation of glutathione (GSH) during peroxidase-catalyzed drug metabolism. p-Phenetidine generates radicals through oxidation, triggering the oxidation of GSH to oxidized glutathione (GSSG) along with significant oxygen consumption. p-Phenetidine can be converted into N-(4-ethoxyphenyl)-p-benzoquinone imine (NEPBQI), which reacts with protein thiols, leading to a decrease in intracellular protein thiols and causing cytotoxicity. p-Phenetidine can be used for drug-induced hepatotoxicity and nephrotoxicity study[1][2].
