Reelin/RELN 蛋白, Human (P.pastoris, His)

Reelin/RELN is a secretory glycoprotein belonging to the extracellular matrix family. Reelin/RELN can bind to ApoER2/VLDLR to activate Src family kinase-mediated Dab1 phosphorylation, initiate PI3K/Akt/mTOR signaling to regulate neuronal migration and synaptic plasticity. Reelin/RELN also activates Rap1 to regulate the actin skeleton, and terminates Reelin/RELN signaling through Cul5/Rbx2/SOCS7 and ADAMTS-4/5. Reelin/RELN abnormalities are closely related to diseases such as lissencephaly caused by RELN mutations, autism, schizophrenia, and Alzheimer's disease. Reelin/RELN Protein, Human (P. pastoris, His) is a recombinant human Reelin/RELN protein expressed from P. pastoris with an N-6*His tag.

Reelin/RELN 激活 Rap1 以调控肌动蛋白细胞骨架重组 (通过 N-cofilin 磷酸化) 和神经元极性，通过下调 E-钙粘蛋白促进上皮-间质转化。Reelin/RELN 信号通过 Dab1 泛素化 (Cul5/Rbx2/SOCS7 复合体) 和 Reelin 蛋白水解 (ADAMTS-4/5、tPA) 终止，或被 ApoE4 通过受体循环缺陷损害。Reelin/RELN 的上游调控因子包括病毒启动子；下游靶点包括 Dab1、PI3K、GSK-3β 及 NMDA/AMPAR 亚基^[1][2]。
Reelin/RELN 突变可能导致无脑回畸形，与自闭症 (标志是脑/血中 Reelin 水平降低) 和精神分裂症 (标志是启动子高甲基化) 相关。杂合 reeler 小鼠可模拟感觉运动缺陷1,5。而 Reelin/RELN 缺乏加速阿兹海默症 (AD) 中 Aβ 斑块形成，而回补 Reelin/RELN 可挽救突触可塑性^[1][2]。
