Resazurin (Diazoresorcinol) sodium, indicator is a non-toxic, stable, membrane-permeable blue non-fluorescent dye (faintly fluorescent). Resazurin sodium, indicator is used as a redox indicator, can be reduced to pink, highly fluorescent Resorufin (Ex=530-560 nm, Em=590 nm) in living cells. Resazurin sodium, indicator can be used for the detection of cell viability, toxicity, proliferation, migration and invasion in cells (human, plant and animal, bacterial and fungal)[1][2][3].

Resazurin sodium, indicator 常用于通过其还原为荧光产物 resorufin 测量细菌和真核细胞的活力。在 TSBc 培养基中，Resazurin sodium, indicator 在浓度为 44 μM 的情况下接种 F. tularensis LVS 后 24 小时未检测到可存活的细菌。将 Resazurin sodium, indicator 的浓度降低至 4.4 μM 仍可导致 F. tularensis LVS 的存活率比单纯培养基减少 10 倍。这两种 Resazurin sodium, indicator 处理在 22 小时内均显著减少了存活的 F. tularensis LVS 细菌数量。在感染 HEK293 细胞 22 小时后，Resazurin sodium, indicator 处理显著减少了可存活的 F. tularensis LVS 细菌数量[1]。

1.溶液配制[2][3]
1.1 原液配制
溶剂：DMSO 或 ddH2O
浓度：1 mg/mL (根据实验优化)。
储存：分装后，-20°C 或 -80°C 避光保存。避免反复冻融。
1.2 工作液配制
用 PBS 或无血清培养基稀释至 1 µg/mL (根据实验优化)。
注意：工作液应立即配制并使用，避光保存。

2. 细胞活力
1. 在 37°C 水浴中解冻 Resazurin sodium, indicator 溶液。
2. 将细胞放在 96 孔板上，用 PBS 清洗（避免光照）。
3. 去除 PBS 清洗液，然后加入 500 μL (1 μg/mL) 的 Resazurin sodium, indicator 溶液。
4. 将板放在培养箱中 30 分钟 (培养时间取决于细胞类型和细胞数量)。
5. 使用微孔板读数仪测量 Resazurin sodium, indicator 荧光 (Ex=530-560 nm，Em=590 nm)。
