AZ3246 is an orally active and selective inhibitor of HPK1 with an IC50 < 3 nM. AZ3246 induces IL-2 secretion in T cells with an EC50 of 90 nM. AZ3246 is a low-clearance antitumor compound[1].

AZ3246 (100 nM) 对 HPK1 的选择性比 STK3、STK4 和 TNIK 激酶高，仅对 HPK1 和 MYLK 有 >80% 的抑制作用，对 ROS1、YSK4 和 TNIK 有 >50% 的抑制作用[1]。
AZ3246 在人肝微粒体中具有出色的代谢稳定性[1]。
AZ3246 诱导 T 细胞分泌 IL-2，EC50 为 90 nM，且不抑制拮抗性激酶。 AZ3246 在原代人、大鼠和狗肝细胞中表现出较低的 CLint 值。在大鼠、狗、猴子和人类肝细胞的 HμREL 共培养中也观察到了低周转率[1]。
