生物活性

(E/Z)-Mirin is a mixture of (E)-Mirin and Mirin ((Z)-Mirin) (HY-19959) configurations. Among them, Mirin is an inhibitor of MRN (Mre11-Rad50-Nbs1). Mirin prevents MRN-dependent ATM activation without affecting ATM protein kinase activity[1][2].

体外研究
(In Vitro)

Mirin 抑制 H2AX 磷酸化，IC50 为 66 μM。Mirin 还能抑制下游靶点 Nbs1 和 Chk2 的 ATM 依赖性磷酸化，以及在 DSB 反应中 ATM 在 Ser1981 位点的 MRN 依赖性自身磷酸化。浓度为 50 μM 和 100 μM 的 Mirin 可诱导显著的 G2 期停滞。Mirin (10-100 μM) 可抑制 TOSA4 细胞中同源性依赖性的 DNA 修复[1]。
BRCA2 缺陷型细胞也对 Mre11 抑制剂 Mirin 表现出超敏反应[2]。
