CD45 Antibody (YA533) is a Rabbit-derived and non-conjugated IgG monoclonal antibody, targeting to CD45.

受体型酪氨酸蛋白磷酸酶 C (PTPRC) 是蛋白酪氨酸磷酸酶 (PTP) 家族的一员，也被称作 CD45, 是一种跨膜糖蛋白。已知 PTPs 是调节多种细胞过程的信号分子，包括细胞生长、分化、有丝分裂和致癌转化。
PTPRC 包含一个胞外结构域、一个跨膜片段和两个串联胞质内催化结构域，因此被归类为受体型 PTP。
PTPRC 已被证明是 T 细胞和 B 细胞抗原受体信号传导的重要调控者，PTPRC 通过与 DPP4 结合，募集和去磷酸化 SKAP1 和 FYN，正向调节 T 细胞的共激活。PTPRC 也使 LYN 去磷酸化，从而调节 LYN 活性。
PTPRC 通过与抗原受体复合物组分的直接相互作用或通过激活抗原受体信号传导所需的各种 Src 家族激酶发挥作用。PTPRC 也抑制 JAK 激酶，因此作为细胞因子受体信号传导的调节因子。
PTPRC 基因有许多选择性剪接的转录物变体，它们编码不同的同种异构体。

• 
  Western blot analysis of extracts from Jurkat(lane 2(20μg) and Jurkat(lane 3(40μg) using CD45 (HY-P80063 Rabbit mAb. Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST for 2 hour at room temperature. The primary antibody (1/1000) and Loading control antibody (Beta Actin, HY-P80438, 1/10000) was used in 5% non-fat milk in TBST at 4°C overnight. Goat Anti-Mouse/Rabbit IgG-HRP Secondary Antibody (1/10000) was used for 1 hour at room temperature.

  Flow cytometric analysis of 1X10^6 Jurkat cells labeling CD45 Antibody (HY-P80063, red). Cells were fixed with 4% paraformaldehyde and permeabilised with 90% methanol. Then stained with the primary antibody at 1/100 dilution for an hour at 4℃. Alexa Fluor® 488-conjugated AffiniPure Goat Anti-Rabbit IgG H&L (HY-P8002) was used as the secondary antibody at 1/1,000 dilution for 30 minutes at 4℃. Rabbit IgG Isotype Control (HY-P80879, blue) was used as the isotype control, cells without incubation with primary antibody were used as the unlabeled control (black).
