IFN-lambda 3/IL-28B  (Ⅲ 型干扰素 3)

IFN-lambda 3 (IL-28B) 是 III 型干扰素家族的成员。IFN-lambda 3 与 IFN-lambda 2 在氨基酸层面上的序列相似性高达 96%。IFN-lambda3 具有抗病毒、抗肿瘤、免疫调节等活性[1]。IFN-lambda 3 通过IFNλ 受体 1 (IFNLR1) 和 IL-10 受体亚基-β (IL-10RB) 的异二聚体受体复合物进行信号传导。当 IFN-lambda 1 与受体复合物结合时,Jak1 和 Tyk2 将被激活,并导致随后的 IFN-λR1 酪氨酸 (胞内结构域,Tyr406 和 Tyr343,Tyr517 位点) 磷酸化,并激活 STAT1 和 STAT2。激活的 STAT1 和 STAT2 会募集 IRF-9 形成三聚体转录因子复合物 (ISGF3)。所形成的 ISGF3 复合物会易位到细胞核,并促进 IFN 刺激基因 (ISG) 的产生,如 IRF7,MX1 和 OAS1[2]。人 IFN-lambda 3 与小鼠氨基酸同源性为 61.98%。

IFN-lambda 3 (IL-28B) is a member of the Type-III interferon family. IFN-lambda 3 has high similar 96% sequence identity with IFN-lambda 2 at the amino acid level. IFN-lambda 3 has antiviral antitumour and immunomodulatory activities[1]. IFN-lambda 3 exhibits biological activities through a heterodimeric receptor complex comprising IFNλ receptor 1 (IFNLR1) and IL-10 receptor subunit-β (IL-10RB). When binding to the receptor complex, Jak1 and Tyk2 will be activated, and leads to subsequent tyrosine phosphorylation of the IFN-λR1 (intracellular domain, Tyr406 and Tyr343, Tyr517), and activation of STAT1 and STAT2. Activated STAT1 and STAT2 together with IRF-9 (p48) form a trimeric transcription factor complex (ISGF3). The formed ISGF3 complexes then translocate to the nucleus and promotes the production of IFN-stimulated genes (ISGs) such as IRF7, MX1, and OAS1[2]. Human IFN-lambda 3 shares 61.98% common aa identity with mouse.
