ADAR1 抗体 (YA1658)

ADAR1 Antibody (YA1658) is a rabbit-derived non-conjugated IgG antibody (Clone NO.: YA1658), targeting ADAR1, with a predicted molecular weight of 136 kDa (observed band size: 150 kDa). ADAR1 Antibody (YA1658) can be used for WB, IHC-P, ICC/IF, FC experiment in human background.

ADAR 是一种催化双链 RNA (dsRNA) 中腺苷 (adenosine) 脱氨转化为肌苷 (inosine) 的酶，这一过程称为 A-to-I RNA 编辑。该酶通过改变密码子影响 mRNA 翻译 (将肌苷识别为鸟苷)，从而改变蛋白质氨基酸序列；同时可影响 pre-mRNA 剪接、RNA 稳定性、病毒 RNA 复制遗传稳定性，以及 microRNA 生成等 RNA 结构依赖性活动。ADAR 能够编辑病毒 (如 HCV、HIV-1) 和细胞 RNA (如 BLCAP、GRIA2、HTR2C 受体)，在特定位点 (如 GRIA2 的 R/G 位点) 或多位点 (超编辑) 发挥作用。其编辑可产生促病毒 (如 HDV、HIV-1) 或抗病毒 (如 HCV) 效应，机制包括编辑依赖性 (HDV) 或非依赖性 (HIV-1 通过抑制 EIF2AK2/PKR) 途径。对 HDV 的编辑可将终止密码子 UAG 变为 UIG，促进大 delta 抗原 (L-HDAg) 合成，但过高 ADAR 活性会抑制 HDV 复制。

• 
  Western blot analysis of extracts from Hela (lane 2(20μg) , HEK293 (lane 3(20μg) and HepG2(lane 4(20μg) using ADAR1 (HY-P81913) Rabbit mAb. Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST for 2 hour at room temperature. The primary antibody (1/1000) and Loading control antibody (Beta Actin, HY-P80438, 1/10000) was used in 5% non-fat milk in TBST at 4°C overnight. Goat Anti-Mouse IgG-HRP Secondary Antibody (1/10000) was used for 1 hour at room temperature.

  Immunocytochemistry analysis of Hela cells labeling ADAR1 with ADAR1 antibody (HY-P81913) at 1/50 dilution. Cells were fixed in 4% paraformaldehyde for 15 minutes at room temperature, permeabilized with 0.1% Triton X-100 for 10 minutes at room temperature, then blocked with QuickBlock™ Blocking Buffer for Immunol Staining for 10 min at room temperature. Cells were then incubated with ADAR1 antibody (HY-P81913) at 1/50 dilution in QuickBlock™ Blocking Buffer for Immunol Staining at 4 ℃. Alexa Fluor® 488-conjugated AffiniPure Goat Anti-Rabbit IgG H&L（HY-P8002, Green) was used as the secondary antibody at 1/1,000 dilution. PBS instead of the primary antibody was used as the secondary antibody only control. The Nuclear counterstain was DAPI (Blue).

  Flow cytometric analysis of 1X10^6 Hela cells labeling ADAR1 Antibody (YA1658) (HY-P81913, red). Cells were fixed with 4% paraformaldehyde and permeabilised with 90% methanol. Then stained with the primary antibody at 1/50 dilution for an hour at 4℃. Alexa Fluor® 488-conjugated AffiniPure Goat Anti-Rabbit IgG H&L (HY-P8002) was used as the secondary antibody at 1/1,000 dilution for 30 minutes at 4℃. Rabbit IgG Isotype Control (HY-P80879, blue) was used as the isotype control, cells without incubation with primary antibody were used as the unlabeled control (black).
