Papain 处理红细胞 ADCC 法[2]
1. 将 Papain 在 37℃ 下孵育 15 min，用磷酸盐缓冲盐水 (PBS) 中洗涤三次。
2. 在微量滴定板中分别评估了单克隆或多克隆抗 D 或抗 A 对酶处理的 O Rh 阳性或 A Rh 阴性红细胞的凝集。
3. 靶点红细胞在 ADCC 检测前用铬酸钠标记。
Note:
活化：木瓜蛋白酶最适 pH 范围为 6.0-7.0，使用前应在活化液中孵育，以确保充分活化。
活化方法：将酶在含有 1.1 mM EDTA，0.067 mM 巯基乙醇和 5.5 mM 半胱氨酸-HCl 的溶液中孵育 30 分钟。
