DDB1 Antibody (YA5274) is a Mouse-derived and non-conjugated monoclonal antibody, targeting to DDB1.
研究背景

DDB1 是一种多功能蛋白质，参与 DNA 修复和蛋白质泛素化过程。作为 UV-DDB 复合物（UV 损伤 DNA 结合蛋白复合物）的核心组分，它识别 UV 诱导的 DNA 损伤（如环丁烷嘧啶二聚体 CPD、6-4 光产物 6-4 PP 等），并招募核苷酸切除修复 NER 通路蛋白启动修复。同时，DDB1 是多种 DCX（DDB1-CUL4-X-box）E3 泛素连接酶复合物的核心组件，通过招募不同底物识别组分（如 DDB2、DTL、ERCC8 等）介导靶蛋白（如组蛋白 H2A/H3/H4、XPC、CRY1 等）的泛素化降解，从而调控 DNA 损伤应答、转录偶联修复 TCR、生物钟功能（如降解 CRY1）等过程。此外，DDB1 通过类似机制（by similarity）参与 FOXO1 蛋白稳定性维持、原始卵泡阶段卵母细胞维持（通过作用于 TET 双加氧酶）以及合子基因组激活等生殖相关功能。
