Polymeric Immunoglobulin Receptor

The polymeric immunoglobulin receptor (pIgR) belongs to the super-Ig family, binds IgM and IgA, and is expressed on the basolateral surface of gut epithelium and in the ducts of secretory glands. PIgR is a highly glycosylated, type I transmembrane protein with a predicted molecular mass of ~81 kDa that is conserved among all vertebrates. The extracellular portion is composed of six domains: five immunoglobulin-like domains, and a sixth, which contains a highly conserved cleavage signal. The intracellular domain contains signals for endocytosis, intracellular sorting and transcytosis. The main function of pIgR is to transport dimeric immunoglobulin A (dIgA) and polymeric immunoglobulin M (pIgM) from the lamina propria across the epithelial barrier to mucosal surfaces in four main steps.
