The EphA3 protein is a receptor tyrosine kinase that promiscuously binds to membrane-bound ephrin ligands to initiate bidirectional signaling. Activation of EphA3 is known to preferentially bind to EFNA5 and regulates cell-cell adhesion, cytoskeletal organization, and migration. The activity was measured by off-chip mobility shift assay(MSA). The enzyme was incubated with fluorecence-labeled substrate and Mg(or Mn)/ATP. The phosphorylated and unphosphorylated substrates were separated and detected by MSA device. When the protein concentration is 5 nM, the conversion rate is about 20%.
