Exodus-2/CCL21 蛋白, Human

Exodus-2/CCL21 Protein, Human is a homeostatic lymphoid chemokine that contributes to the entry of T cells and dendritic cells into the lymphoid T-zone. It acts through chemokine receptors CCR7 and CXCR3 to promote fibrogenic and inflammatory cytokine production. Exodus-2/CCL21 Protein, Human  is a recombinant human Exodus-2/CCL21 (S24-P134) expressed by E. coli^[1].

CCL21，也称为 exodus-2 和次级淋巴趋化因子 (SLC)，是属于 CC 趋化因子家族的一种小细胞因子，位于人类基因组中的第 9 号染色体上。它与糖胺聚糖 (GAG) 结合并固定在内皮细胞的表面。作为一种趋化因子，CCL21 抑制造血并刺激趋化性，并且在体外对胸腺细胞和活化的 T 细胞具有趋化性，但对 B 细胞、巨噬细胞或中性粒细胞没有趋化性。同时，CCL21是T细胞迁移和粘附的强效刺激剂，与T细胞上的糖蛋白PSGL-1结合，促进T细胞迁移至次级淋巴器官。CCL21 可通过趋化因子受体 CCR7 和 CXCR3 发挥作用。其中，CCR7是一种GPCR，通常由T细胞亚群中央记忆细胞、胸腺T细胞、B细胞、成熟DC和其他稀有细胞亚群表达。ccl21 可作为中枢神经系统中的小胶质细胞激活剂，仅在濒危或机械损伤的神经元中表达^[1][2]。

Namru 乳腺 (NMuMG) 上皮细胞经历 TGF-β1（10 ng/mL，48 小时）诱导的上皮间质转化 (EMT) 后，获得向 CCL21 (350 ng/mL；48 h) 迁移的能力。TGF-β1 促进 CCR7 阳性免疫细胞的趋化迁移^[5]。
CCL21 (50 ng/mL, 100 ng/mL, 200 ng/mL;48 h) 以浓度依赖性方式促进 T24 细胞增殖，在 200 ng/mL 浓度下效果显著^[6]。

CCL21(0.1 µg/mL, 2 h) can interact with polysialic acid to regulate the migratory capacity of human dendritic cells^[3].

It is not recommended to reconstitute to a concentration less than 100 μg/mL in ddH₂O. For long term storage it is recommended to add a carrier protein (0.1% BSA, 5% HSA, 10% FBS or 5% Trehalose).

Stored at -20°C for 2 years. After reconstitution, it is stable at 4°C for 1 week or -20°C for longer (with carrier protein). It is recommended to freeze aliquots at -20°C or -80°C for extended storage.
