MDC/CCL22 蛋白, Mouse (His)

CCL22/MDC Protein,Mouse (His) is a CC chemokine that acts as a ligand for the CCR4 receptor and is a chemoattractant for CCR4-expressing cells such as Th2 cells, playing an important role in homeostatic and inflammatory responses. CCL22/MDC Protein, Mouse (His) is a recombinant mouse CCL22/MDC (G26-S92) protein expressed by E. coli with a his tag^[1][2].

CCL22又称巨噬细胞源性趋化因子（MDC），是一种位于人类基因组16号染色体上的CC趋化因子，是由CCL22基因编码的蛋白质，在氨基酸水平上与CCL17有37%的同一性。CCL22由树突状细胞和巨噬细胞分泌，可被脂多糖、细胞因子等多种刺激因子上调。其中，Th2细胞因子IL-4和IL-13诱导骨髓细胞产生CCL22，并可被Th1细胞因子IFN-γ抑制。除了作为表达 CCR4 的 Th2 淋巴细胞、单核细胞、单核细胞衍生的树突细胞和自然杀伤细胞的有效趋化剂外，CCL22 还可以通过与趋化因子受体 CCR4 相互作用来影响其靶细胞。CCL22是CCR4内化的有效诱导剂，CCL22与CCR4的结合降低了CCR4随后的功能反应。CCL22 与 CCR4 的相互作用涉及多种病理学，从过敏反应、自身免疫到肿瘤生长。相比之下，能够阻断 CCL17 和 CCL22 介导的 Th2 和 Treg 细胞募集的小分子化合物和抗体已被证明在哮喘、特应性疾病和肿瘤生长的各种疾病模型中具有积极作用。此外，CCL22 及其受体在 TH2 淋巴细胞募集中的重要作用已在过敏性气道炎症模型中得到证实。它还可以通过调节成熟胸腺细胞通过该器官的迁移来参与胸腺生成^[1][2]。

CCL22 (1 and 1 ng/mL, 4 h) is an effective stimulator of eosinophil degranulation, but is ineffective for eosinophil chemotaxis^[3].

CCL22 (intrapleurally (i.pl.), 1-1 ng/cavity) can cause recruitment of eosinophils into the pleural cavity of mice in a dose- and time-dependent manner^[3].
CCL22/MDC ( intravenously, 1μg) increases lung inflammation in male C57BL/6 mice, but does not lead to increased lung cell recruitment in normal (i.e., non-hemorrhagic) mice. This suggests that MDC plays an important role in the pathogenesis of acute lung inflammation in the context of hemorrhage and resuscitation^[4].
