MT-RNR1 通过抑制 folate cycle 减少 de novo purine 生物合成，导致中间产物 5-aminoimidazole-4-carboxamide (AICAR) 积累并激活代谢调节因子 AMPK，从而调控胰岛素敏感性和代谢稳态。它能抵抗年龄和饮食诱导的胰岛素抵抗及肥胖。在代谢应激时，MT-RNR1 转位至细胞核，结合抗氧化反应元件 (ARE) 并通过 NFE2L2 依赖的方式调节核基因表达，增强细胞对代谢应激的抵抗。此外，它增加衰老细胞中线粒体呼吸及 CPT1A 和细胞因子 (IL1B、IL6、IL8、IL10、TNF) 水平。MT-RNR1 还通过激活 mTORC2 和抑制 PTEN 磷酸酶活性促进 AKT 磷酸化，进而抑制 FOXO1 活性并减少 MSTN 表达，促进骨骼肌生长。通过 TGFB/SMAD 通路，它促进骨髓间充质干细胞的成骨分化，并刺激成骨细胞增殖及 I 型胶原 (COL1A1、COL1A2) 合成。
