DNA Ligase IV Antibody (YA2699) is a Rabbit-derived and non-conjugated IgG monoclonal antibody, targeting to DNA Ligase IV.

DNA Ligase IV 是一种参与 DNA 非同源末端连接 (NHEJ) 的 DNA 连接酶，在双链断裂 (DSB) 修复和 V(D)J 重组中发挥关键作用。它通过重新密封 DNA 断裂来完成 DSB 修复中的 NHEJ 连接步骤，并能在 ATP 依赖的反应中连接双链多聚脱氧核苷酸中的单链断裂。LIG4 具有机制灵活性：可以单独连接切口和兼容的 DNA 突出端，而在 XRCC4 存在的情况下，还能连接具有 2 个核苷酸 (nt) 微同源性和 1-nt 缺口的末端。LIG4 与 XRCC4 形成亚复合物，LIG4-XRCC4 亚复合物负责 NHEJ 连接步骤，且 XRCC4 增强 LIG4 的连接活性。LIG4-XRCC4 复合物与 DNA 末端的结合依赖于 DNA 依赖性蛋白激酶复合物 DNA-PK 在这些 DNA 末端的组装。此外，LIG4 还调节 XRCC4 的核定位。

  Western blot analysis was performed on extracts from Hela (lane 1, 15 μg), Jurkat (lane 2, 15 μg), Ramos (lane 3, 15 μg), HepG2 (lane 4, 15 μg), and U251 (lane 5, 15 μg) using DNA Ligase IV Rabbit mAb.Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST at 4°C overnight.The primary antibody (1:1000 dilution) and the loading control antibody (beta-Tubulin, HY-P80487, 1:20000 dilution) were incubated in 5% non-fat milk in TBST for 1 hour at 37°C.Goat Anti-Rabbit IgG-HRP Secondary Antibody (1:20000 dilution) was then applied for 40 minutes at 37°C.
