      描述及优势

      MCE 特超敏 ECL 化学发光试剂盒 (Ultra High Sensitive ECL Kit) 应用检测辣根过氧化物酶 (HRP) 标记的抗体及其关联的抗原。HRP 标记的抗体直接或间接结合膜上的目的蛋白，在洗膜后加入本试剂盒配制的 ECL 工作液，即可由 HRP 催化发出荧光。结果可以通过 X 光胶片压片或者其他荧光成像设备进行检测。 本产品配方独特，灵敏度极高，对抗原的最低检测限可达中飞克级别，在western blot 实验中，一抗 (1 mg/mL) 储存液可稀释1:1000 至1:50000 倍，二抗 (1 mg/mL) 储存液可稀释 1:50000 至 1:200000 倍。持久的发光信号，极高的信噪比，保证了最佳的检测效果。

      操作流程

      1. 将 MCE 特超敏 ECL 化学发光试剂盒 (Ultra High Sensitive ECL Kit) 从冰箱中取出，放置在室温下平衡约 20 分钟。

      2. 配制 ECL 工作液：按等体积比例混合适量 A 液和 B 液，室温放置备用。工作液最好在临近使用前配制。

      3. 根据印迹膜的大小，按 0.1 mL 工作液/cm2 膜的比例滴加 ECL 工作液到膜上，确保工作液均匀覆盖在整张印迹膜上。

      4. 室温孵育 1-2 分钟。

      5. 弃去印迹膜上的 ECL 工作液，用吸水纸略吸去过多的液体。将印迹膜放在两片保鲜膜中间，随后进行压片检测或荧光成像仪检测。

      6. 压片检测：将印迹膜固定于片夹内。暗室内压片 1 分钟，立即显影定影，根据结果再调整压片时间。或直接压片 30 秒、1、3、5 分钟，然后一起显影定影观察结果。

      7. 荧光成像仪检测：将印迹膜放置到荧光成像仪内，参考仪器说明书进行检测。

      • Do most proteins show cross-species activity?

        Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.
