GLUL 是一种 glutamine synthetase，能够催化 ATP 依赖的 glutamate 和 ammonia 转化为 glutamine。其功能因组织定位而异：在大脑中，它调节有毒 ammonia 的水平并将神经毒性 glutamate 转化为无害的 glutamine ；而在肝脏中，它是负责清除 ammonia 的酶之一（ by similar ）。在红细胞生成过程中，GLUL 在铵解毒中起关键作用：其 glutamine synthetase 活性用于清除 heme 生物合成过程中 porphobilinogen deaminase (HMBS) 产生的铵，以防止铵积累和氧化应激（ by similar ）。此外，GLUL 对胎儿皮肤成纤维细胞的增殖至关重要。独立于其 glutamine synthetase 活性，GLUL 在血管发育过程中调节内皮细胞迁移，通过调控 GTPase RHOJ 的膜定位和激活（可能通过促进 RHOJ palmitoylation ）发挥作用。它可能作为 RHOJ 的 palmitoyltransferase，能够先 autopalmitoylate，然后将 palmitoyl 基团转移至 RHOJ。GLUL 还参与 ribosomal 40S subunit 的生物合成，并通过与 BEST2 的相互作用调节其通道活性：在无细胞内 L-glutamate 时抑制 BEST2 的开放，但在存在 L-glutamate 时促进 BEST2 的开放，从而缓解其对 BEST2 的抑制效应。
