RUNX1 与 CBFB 形成异二聚体复合物 core-binding factor (CBF)。RUNX 家族成员通过其 runt 结构域识别调控区域中的核心结合序列 5'-TGTGGT-3' 或极少数情况下的 5'-TGCGGT-3'，从而调控靶基因的转录，而 CBFB 是一种非 DNA 结合的调节亚基，可通过变构效应增强 RUNX 的序列特异性 DNA 结合能力。该异二聚体结合多种增强子和启动子的核心位点，包括鼠白血病病毒、多瘤病毒增强子、T 细胞受体增强子、LCK、IL3 和 GM-CSF 启动子。RUNX1 对正常造血功能的发育至关重要，与 ELF4 协同激活 IL-3 启动子，与 ELF2 协同激活 BLK 启动子。它抑制 KAT6B 依赖的转录激活 (by similarity)，并参与未成熟 T 细胞前体的谱系定向。CBF 复合物在细胞毒性 (CD8+) T 细胞发育过程中抑制 ZBTB7B 转录因子，通过结合 ZBTB7B 基因座的 RUNX 结合序列作为转录沉默子，促进细胞毒性 T 细胞分化。此外，RUNX1 通过结合 FOXP3 调控调节性 T 细胞 (Treg) 的无能和抑制功能，在常规 T 细胞中激活 IL2 和 IFNG 表达，下调 TNFRSF18、IL2RA 和 CTLA4 表达 (by similarity)。在 T-helper 17 细胞中，RUNX1 正向调控 RORC 表达。Isoform AML-1G 对靶基因的结合活性更高，与 TCR-beta-E2 和 RAG-1 靶位点的结合亲和力是其他亚型的三倍，但在中性粒细胞终末分化中效果较弱。Isoform AML-1L 干扰 RUNX1 的反式激活活性。

  Immunohistochemical analysis of paraffin-embedded Mouse colon tissue using RUNX Antibody. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0) for 2 minutes. The tissues were blocked in QuickBlock for 20 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody (HY-P80888, 1/100) in 4℃ overnight. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.
