3,5-Dinitrosalicylic acid the derivative of salicylic acid. 3,5-Dinitrosalicylic acid is used in the α-amylase assay, carbohydrase assay, and for the colorimetric determination of reducing substances[1].

3,5-Dinitrosalicylic acid 是一种常用的还原糖检测试剂，3,5-Dinitrosalicylic acid 与还原糖发生氧化还原反应生成衍生物，从而产生颜色变化，用于定量检测还原糖含量。
配制两种不同的 DNSA 试剂：
1.试剂配制：
DNSA-1 试剂：0.4M NaOH 溶液，每 100mL 溶液中含有一定量的七水合物。
DNSA-2 试剂：0.7M NaOH 溶液，每 100mL 含有 2g 3,5-Dinitrosalicylic acid。
2.样品配制：
配制一系列不同浓度（0.15 - 5.85mM）的麦芽糊精溶液。若研究钙离子的作用，则在麦芽糊精溶液中加入氯化钙至浓度为 40mM。
3.测试步骤：
取 2mL 样品置于 Folin-Wu 糖管中，加入 DNSA 试剂，在沸水浴中加热 5 min，再在冷流水中冷却 5 min，冷却后稀释至 25mL，用带绿色滤光片的 Unicam SP 1300 比色计测量吸光度。
4.结果分析：
以吸光度（A）为纵坐标，麦芽糊精的毫摩尔浓度（M）为横坐标绘制曲线，通过绘制每种麦芽糊精的吸光度-浓度曲线的斜率与麦芽糊精的大小（葡萄糖残基数）的关系，比较不同麦芽糊精的摩尔减少值。若试剂的吸光度值与还原端数目成正比，则曲线的斜率为零；若吸光度值与还原端数目不成正比，且随麦芽糊精粒径的增大而增加，则曲线的斜率大于零。
