6-AF488 tyramide is a bright, green fluorescent dye (Ex=496 nm, Em=524 nm). 6-AF488 tyramide is utilized as reporter fluorescent substrate of horseradish peroxidase (HRP)-catalyzed deposition for tyramide signal amplification (TSA). 5-FITC tyramide can be used for multiplex immunohistochemistry (mIHC)[1].

体外研究
(In Vitro)

6-AF488 tyramide 检测[1]。
1) 将组织切片放入 24 孔板的每个孔中，切勿让组织部分干燥。
2) 用含 0.2% Triton X-100 的 PBS 溶液通透组织 5-10 分钟。
3) 用 PBS-T 冲洗 5-10 分钟，然后将组织放入 1% H2O2 溶液中孵育 20 分钟，以阻断内源性过氧化物酶活性。
4) 用 PBS 冲洗三次，每次 5-10 分钟，然后将组织放入含 1% 封闭液 (4% 正常驴血清+ PBS-T) 的 PBS 溶液中孵育 1 小时，以阻断非特异性结合位点。
5) 加入一抗工作液室温孵育 2 小时或 4℃ 过夜，然后用 PBS 冲洗 3 次，每次 5-10 分钟。
6) 加入 HRP 标记的羊抗兔二抗 (1: 100 溶于 1% 封闭液中) 室温孵育 2 小时，然后用 PBS 冲洗 3 次，每次 5-10 分钟。
7) 用 0.0015% H2O2 激活 TSA 缓冲液，然后将组织放入 6-AF488 tyramide 工作液 (1: 100 溶于激活的 TSA 缓冲液中) 中室温孵育 5-10 分钟。
8) 用 PBS 冲洗 3 次，每次 5-10 分钟，然后将切片固定到载玻片上。
9) 将载玻片以近乎垂直的位置放入载玻片支架中，静置 10 分钟使其干燥，再用 ddH2O 快速冲洗载玻片。
10) 滴加封片剂封片，我们推荐使用 AntiFade Mounting Medium (with DAPI) (HY-K1047)。
11) 荧光显微镜检测。
注：建议根据实际情况确定 6-AF488 tyramide 工作液的最佳浓度。
