7-Methyl-6-thioguanosine (MESG) is a chromogenic substrate that can be converted to 7-methyl-6-thioguanine in the presence of purine nucleoside phosphorylase and inorganic phosphate. 7-Methyl-6-thioguanosine can be used to quantify inorganic phosphate. 7-Methyl-6-thioguanosine can also be used to determine the activities of purine nucleoside phosphorylase and protein phosphatases[1][2].

测定蛋白磷酸酶简要步骤[1]:
1. 试剂准备: 将 7-Methyl-6-thioguanosine 用 DMSO 配制成储备液，当天使用，通过 331nm 处吸光值及对应摩尔消光系数确定其浓度。
2. 配置反应体系: 依据检测的蛋白质磷酸酶类型配置反应混合物。
3. 反应检测: 25°C 平衡反应混合物 5 分钟后加入蛋白质磷酸酶启动反应，用双光束分光光度计在 360nm 记录吸光度变化，通过软件分析反应曲线斜率确定初始反应速率，依据特定消光系数定量释放的磷酸盐。
4. 数据分析: 用非线性回归分析程序处理数据，计算 Km、Vmax 等参数评估蛋白质磷酸酶活性。
