Aconitase 2 Antibody (YA2151) is a Rabbit-derived and non-conjugated IgG monoclonal antibody, targeting to Aconitase 2.
研究背景

Aconitase 2 是一种含 [4Fe-4S] 簇的铁硫酶（iron-sulfur enzyme），通过顺乌头酸（cis-aconitate）中间体催化柠檬酸（citrate）与异柠檬酸（isocitrate）的相互转化，在三羧酸循环（TCA cycle）和乙醛酸循环（glyoxylate cycle）中起关键作用。该酶对氧化损伤高度敏感，其 [4Fe-4S] 簇易被超氧阴离子（superoxide）、H2O2 和 NO 等活性氧/氮物种破坏，导致铁离子释放并加剧细胞氧化应激。Aconitase 2 的活性受镁离子浓度和柠檬酸/异柠檬酸比例调控，硫氧还蛋白（thioredoxin）可修复其氧化损伤后的活性中心。

  • Western blot analysis was performed on extracts from Hela (lane 1, 15 μg), K562 (lane 2, 15 μg), Jurkat (lane 3, 15 μg), HepG2 (lane 4, 15 μg), Mouse liver (lane 5, 15 μg) , Rat liver (lane 5, 15 μg) using Aconitase 2 Rabbit mAb.Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST at 4°C overnight.The primary antibody (1:2000 dilution) and the loading control antibody (beta-Actin, HY-P83730, 1:20000 dilution) were incubated in 5% non-fat milk in TBST for 1 hour at 37°C.Goat Anti-Rabbit IgG-HRP Secondary Antibody (1:20000 dilution) was then applied for 40 minutes at 37°C.
