ALKBH4 Antibody (YA2050) is a Rabbit-derived and non-conjugated IgG monoclonal antibody, targeting to ALKBH4.

ALKBH4 是一种双加氧酶（dioxygenase），能够介导肌动蛋白（actin）单甲基化位点 ‘Lys-84’（K84me1）的去甲基化，从而调控肌动球蛋白（actomyosin）相关过程（by similar）。该酶对肌动蛋白 K84me1 的去甲基化作用对于维持细胞分裂（cytokinesis）和细胞迁移（cell migration）过程中正常的收缩环（cleavage furrow）内陷所需的肌动球蛋白动力学至关重要（by similar）。除蛋白质外，ALKBH4 还能对 DNA 进行去甲基化，特异性作用于腺嘌呤第 6 位甲基化（N(6)-methyladenosine）的 DNA，从而参与调控 Polycomb 沉默（Polycomb silencing）机制（by similar）。

  Western blot analysis was performed on extracts from 293 (lane 1, 20 μg), MOLT-4 (lane 2, 20 μg), K562 (lane 3, 20 μg), and HeLa (lane 4, 20 μg) using ALKBH4 Rabbit mAb.Proteins were transferred to a PVDF membrane and blocked with 5% non - fat milk in TBST at 4°C overnight.The primary antibody (1:1000 dilution) and the loading control antibody (beta-Tubulin, HY-P80955, 1:5000 dilution) were incubated in 5% non-fat milk in TBST for 1 hour at 37°C.Goat Anti - Rabbit/Mouse IgG - HRP Secondary Antibody (1:20000 dilution) was then applied for 40 minutes at 37°C.
