CD31 Antibody (YA541) is a Rabbit-derived and non-conjugated IgG monoclonal antibody, targeting to CD31.

CD31 是一种细胞粘附分子，在大多数炎症条件下参与白细胞跨内皮迁移 (TEM) 过程。其 Tyr-690 位点对 TEM 至关重要，影响 PECAM1 向内皮细胞侧缘循环区室 (LBRC) 的转运效率，并促进 LBRC 膜围绕迁移白细胞的定向移动。CD31 通过同型互作参与内皮细胞间的连接粘附，与 CD177 的异型互作则影响中性粒细胞的跨内皮迁移。同型结合可传递脱离信号，阻止巨噬细胞对存活白细胞的吞噬，但对凋亡白细胞无此作用，反而促进其被巨噬细胞吞噬。此外，CD31 调节缓激肽受体 BDKRB2 的激活，影响缓激肽和高渗休克诱导的内皮细胞 ERK1/2 信号通路激活 (类似功能提示其可能增加动脉粥样硬化易感性，但无抗凋亡作用)。

  Western blot analysis of extracts from THP-1(lane 2(20μg) and THP-1(lane 3(40μg) using CD31 (HY-P80447) Rabbit mAb. Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST for 2 hour at room temperature. The primary antibody (1/1000) and Loading control antibody (Beta Actin, HY-P80438, 1/10000) was used in 5% non-fat milk in TBST at 4°C overnight. Goat Anti-Mouse/Rabbit IgG-HRP Secondary Antibody (1/10000) was used for 1 hour at room temperature.

  Flow cytometric analysis of 1X10^6 THP-1 cells labeling CD31 (HY-P80447, red). Cells were fixed with 4% paraformaldehyde and permeabilised with 90% methanol. Then stained with the primary antibody at 1/50 dilution for an hour at 4℃. Alexa Fluor® 488-conjugated AffiniPure Goat Anti-Rabbit IgG H&L (HY-P8002) was used as the secondary antibody at 1/1,000 dilution for 30 minutes at 4℃. Rabbit IgG Isotype Control (HY-P80879, blue) was used as the isotype control, cells without incubation with primary antibody were used as the unlabeled control (black).
