CRM1 Antibody (YA1554) is a Rabbit-derived and non-conjugated IgG monoclonal antibody, targeting to CRM1.

CRM1 是一种介导细胞蛋白质（含富含亮氨酸的核输出信号 NES）和 RNA 核输出的蛋白质。在细胞核中，它与 RANBP3 协同结合靶蛋白的 NES 和 GTP 酶 RAN 的活性 GTP 结合形式（Ran-GTP）。该复合物通过与核孔复合体（NPC）中的核孔蛋白结合，锚定在核孔上。当核输出复合物转运至细胞质后，复合物解离以及 Ran-GTP 水解为 Ran-GDP（分别由 RANBP1 和 RANGAP1 诱导）导致货物从输出受体释放。核输出的方向性被认为是由 Ran 的 GTP 和 GDP 结合形式在细胞质和细胞核中的不对称分布决定的。此外，CRM1 参与 U3 snoRNA 从 Cajal 体到核仁的运输，并结合带有 TMG 帽的晚期前体 U3 snoRNA；（微生物感染）它还介导多种病毒（包括 HIV-1、HTLV-1 和甲型流感病毒）未剪接或不完全剪接 RNA 的核输出，并与 HIV-1 Rev 和 HTLV-1 Rex 蛋白相互作用，介导它们的核输出，同时参与 HTLV-1 Rex 的多聚化。

• 
  Western blot analysis of extracts from Hela(lane 2(20ug) , PANC-1(lane 3(20ug) and NIH/3T3(lane 4(20ug) using CRM1 Antibody (HY-P81809) Rabbit mAb. Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST for 2 hour at room temperature. The primary antibody (1/1000) and Loading control antibody (Beta Actin, HY-P80438, 1/10000) was used in 5% non-fat milk in TBST at 4°C overnight. Goat Anti-Mouse/Rabbit IgG-HRP Secondary Antibody (1/10000) was used for 1 hour at room temperature.

  Immunocytochemistry analysis of Hela cells labeling CRM1 with CRM1 Antibody (HY-P81809)at 1/50 dilution. Cells were fixed in 4% paraformaldehyde for 15 minutes at room temperature, permeabilized with 0.1% Triton X-100 for 10 minutes at room temperature, then blocked with QuickBlock™ Blocking Buffer for Immunol Staining for 10 min at room temperature. Cells were then incubated with CRM1 Antibody (HY-P81809) at 1/50 dilution in QuickBlock™ Blocking Buffer for Immunol Staining at 4 ℃. Alexa Fluor® 488-conjugated AffiniPure Goat Anti-Rabbit IgG H&L（HY-P8002, Green) was used as the secondary antibody at 1/1,000 dilution. PBS instead of the primary antibody was used as the secondary antibody only control. The Nuclear counterstain was DAPI (Blue).

  Flow cytometric analysis of 1X10^6 Hela cells labeling CRM1 Antibody (YA1554) (HY-P81809, red). Cells were fixed with 4% paraformaldehyde and permeabilised with 90% methanol. Then stained with the primary antibody at 1/50 dilution for an hour at 4℃. Alexa Fluor® 488-conjugated AffiniPure Goat Anti-Rabbit IgG H&L (HY-P8002) was used as the secondary antibody at 1/1,000 dilution for 30 minutes at 4℃. Rabbit IgG Isotype Control (HY-P80879, blue) was used as the isotype control, cells without incubation with primary antibody were used as the unlabeled control (black).
