GCN2 Antibody (YA417) is a Rabbit-derived and non-conjugated IgG monoclonal antibody, targeting to GCN2.

GCN2 是一种代谢应激感应蛋白激酶，能够在氨基酸可用性低时磷酸化真核翻译起始因子 2 的 α 亚基 (EIF2S1/eIF-2-alpha)，从而激活整合应激反应 (ISR) 以适应氨基酸饥饿 (By similar)。其磷酸化 EIF2S1/eIF-2-alpha 会抑制全局帽依赖性翻译，同时优先翻译 ISR 特异性 mRNA（如转录激活因子 ATF4），从而通过 ATF4 介导的氨基酸生物合成基因表达重编程缓解营养缺乏 (PubMed:32610081)。此外，GCN2 可结合未充电的 tRNA (By similar)，并在氨基酸饥饿后促进蛋白激酶 PRKCH 的翻译诱导 (By similar)。它还参与细胞周期调控，通过未折叠蛋白反应 (UPR) 激活后抑制 cyclin D1 mRNA 翻译，或在氨基酸剥夺时激活细胞周期抑制剂 CDKN1A/p21 mRNA 翻译 (PubMed:26102367)。在神经系统中，GCN2 影响突触可塑性巩固、学习及长期记忆形成 (By similar)，并抑制神经突生长 (By similar)。在葡萄糖剥夺时发挥促凋亡作用 (By similar)，且能独立于 SAPK/JNK 和 p38 MAPK 信号通路在紫外线照射后抑制全局蛋白质合成 (By similar)。此外，GCN2 通过抑制甲病毒早期病毒 mRNA 翻译发挥抗病毒作用 (By similar)；在黄热病毒感染中，它通过促进树突细胞自噬和抗原呈递调节适应性免疫反应 (PubMed:24310610)。

• 
  Western blot analysis of extracts from A549 (lane 2(20μg) and A549(lane 3(40μg) using GCN2 (HY-P80138) Rabbit mAb. Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST for 2 hour at room temperature. The primary antibody (1/1000) and Loading control antibody (Beta Actin, HY-P80438, 1/10000) was used in 5% non-fat milk in TBST at 4°C overnight. Goat Anti-Mouse/Rabbit IgG-HRP Secondary Antibody (1/10000) was used for 1 hour at room temperature.

  Immunocytochemistry analysis of Hela cells labeling GCN2 with GCN2 Antibody (HY-P80138)at 1/50 dilution. Cells were fixed in 4% paraformaldehyde for 15 minutes at room temperature, permeabilized with 0.1% Triton X-100 for 10 minutes at room temperature, then blocked with QuickBlock™ Blocking Buffer for Immunol Staining for 10 min at room temperature. Cells were then incubated with GCN2 Antibody (HY-P80138) at 1/50 dilution in QuickBlock™ Blocking Buffer for Immunol Staining at 4 ℃. Alexa Fluor® 488-conjugated AffiniPure Goat Anti-Rabbit IgG H&L（HY-P8002, Green) was used as the secondary antibody at 1/1,000 dilution. PBS instead of the primary antibody was used as the secondary antibody only control. The Nuclear counterstain was DAPI (Blue).

  Flow cytometric analysis of 1X10^6 MCF-7 cells labeling GCN2 Antibody(HY-P80138, red). Cells were fixed with 4% paraformaldehyde and permeabilised with 90% methanol. Then stained with the primary antibody at 1/50 dilution for an hour at 4℃. Alexa Fluor® 488-conjugated AffiniPure Goat Anti-Rabbit IgG H&L (HY-P8002) was used as the secondary antibody at 1/1,000 dilution for 30 minutes at 4℃. Rabbit IgG Isotype Control (HY-P80879, blue) was used as the isotype control, cells without incubation with primary antibody were used as the unlabeled control (black).
