Osteocalcin Antibody is a Rabbit-derived and non-conjugated IgG polyclonal antibody, targeting to Osteocalcin.
研究背景

Osteocalcin 是一种骨蛋白，占骨总蛋白的 1-2%，作为骨形成的负调节因子发挥作用。它能够在不影响骨吸收或矿化的情况下限制骨形成（By similarity），并强烈结合磷灰石和钙。未羧化形式的 Osteocalcin 作为由成骨细胞分泌的激素，调节多种细胞过程，如能量代谢、男性生育能力和大脑发育。通过促进胰腺 β 细胞增殖、胰岛素分泌和敏感性以及能量消耗，调节能量代谢。未羧化的 Osteocalcin 还促进睾丸中睾酮的产生：作为 Leydig 细胞表面 G 蛋白偶联受体 GPRC6A 的配体，启动信号传导反应，以 CREB 依赖性方式促进睾酮合成所需酶的表达。此外，Osteocalcin 还调节大脑发育：它穿过血脑屏障，作为神经元上 GPR158 的配体，启动信号传导反应，防止海马神经元凋亡，促进所有单胺类神经递质的合成并抑制 γ-氨基丁酸（GABA）的合成。Osteocalcin 在妊娠期间还能穿过胎盘，母体 Osteocalcin 对胎儿大脑发育至关重要。

  • Western blot analysis of extracts from M-muscle (lane 1) and K562 (lane 2) and M-lung (lane 3) and M-lymph (lane 4) using Osteocalcin antibody. Proteins were transferred to a PVDF membrane and blocked with 5% nonfat powdered milk in PBST for 2 hour at room temperature. The primary antibody (1/1000) and Loading control antibody (GAPDH, 1/3000) was diluted with 5% nonfat powdered milk in PBST at 4°C overnight. Goat Anti-Rabbit IgG-HRP Secondary Antibody (1/8,000) was incubated for 45min at room temperature.
