cAMP is a signaling molecule important for a variety of cellular functions. cAMP exerts its effects by activating the cAMP-dependent protein kinase, which transduces the signal through phosphorylation of different target proteins. The inactive kinase holoenzyme is a tetramer composed of two regulatory and two catalytic subunits. cAMP causes the dissociation of the inactive holoenzyme into a dimer of regulatory subunits bound to four cAMP and two free monomeric catalytic subunits. Four different regulatory subunits and three catalytic subunits have been identified in humans. The related gene encodes one of the regulatory subunits. The protein was found to be a tissue-specific extinguisher that down-regulates the expression of seven liver related genes in hepatoma x fibroblast hybrids. Mutations in The related gene cause Carney complex (CNC). The related gene can fuse to the RET protooncorelated gene by related gene rearrangement and form the thyroid tumor-specific chimeric oncorelated gene known as PTC2. A nonconventional nuclear localization sequence (NLS) has been found for The protein which suggests a role in DNA replication via the protein serving as a nuclear transport protein for the second subunit of the Replication Factor C (RFC4). Several alternatively spliced transcript variants encoding two different isoforms have been observed.

  Western blot analysis was performed on extracts from Hela (lane 1, 15 μg), Rat brain (lane 2, 15 μg), C6 (lane 3, 15 μg), Jurkat (lane 4, 15 μg), and U-87MG (lane 5, 15 μg) using PKA RI alpha Rabbit mAb.Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST at 4°C overnight.The primary antibody (1:1000 dilution) and the loading control antibody (GAPDH, HY-P80137, 1:60000 dilution) were incubated in 5% non-fat milk in TBST for 1 hour at 37°C.Goat Anti-Rabbit IgG-HRP Secondary Antibody (1:20000 dilution) was then applied for 40 minutes at 37°C.
