PUM2 Antibody (YA2586) is a Rabbit-derived and non-conjugated IgG monoclonal antibody, targeting to PUM2.

PUM2 是一种序列特异性 RNA 结合蛋白，通过结合 mRNA 靶标的 3'-UTR 作为转录后抑制因子发挥作用。它结合到 RNA 共有序列 Pumilio Response Element (PRE) 5'-UGUANAUA-3'，该序列与 Nanos Response Element (NRE) 相关。PUM2 通过不同机制介导转录后抑制：通过直接招募 CCR4-POP2-NOT 去腺苷酸酶导致翻译抑制和 mRNA 降解；或通过促进 miRNA 的可及性实现去腺苷酸化非依赖性抑制。PUM2 通过结合 E2F3 mRNA 的 3'-UTR 并促进 miRNA 调控来抑制其表达。此外，它在病毒感染的胞质感知中发挥作用，并抑制维持基因组稳定性所需的基因程序（如关键的有丝分裂、DNA 修复和 DNA 复制因子）。其抑制能力受 lncRNA NORAD (non-coding RNA activated by DNA damage) 调控，NORAD 通过大量 PUMILIO 结合位点隔离 PUM1 和 PUM2。PUM2 可能调控 DCUN1D3 mRNA 水平，并支持干细胞的增殖和自我更新。与 PUM1 共同特异性结合 miRNA MIR199A 前体，在转录后水平调控其表达。

  Western blot analysis of extracts from HEK293T (lane 2(20μg), HEK293T (lane 3(40μg), using PUM2 Antibody. Proteins were transferred to a PVDF membrane and blocked with 5% BSA in TBST for 2 hour at room temperature. The primary antibody and Loading control antibody (Beta Actin, HY-P80438, 1/3000) was used in 5% BSA in TBST at 4°C overnight. Goat Anti-Mouse/Rabbit IgG-HRP Secondary Antibody (HY-P8004/HY-P8001, 1/10,000) was used for 1 hour at room temperature.

  Flow cytometric analysis of 1X10^6 Hela cells labeling PUM2 Antibody (YA2586) (HY-P82841, red). Cells were fixed with 4% paraformaldehyde and permeabilised with 90% methanol. Then stained with the primary antibody at 1/50 dilution for an hour at 4℃. Alexa Fluor® 488-conjugated AffiniPure Goat Anti-Rabbit IgG H&L (HY-P8002) was used as the secondary antibody at 1/1,000 dilution for 30 minutes at 4℃. Rabbit IgG Isotype Control (HY-P80879, blue) was used as the isotype control, cells without incubation with primary antibody were used as the unlabeled control (black).
