CellTracker CM-DiI  (Synonyms: CM-DiI)

CellTracker CM-DiI (CM-DiI) is a lipophilic fluorochrome that can be used for tracking the migration of lymphocytes. CellTracker CM-DiI is feasible to detect labeled cells in histological sections (excitation 553 nm; emission 570 nm)^[1][2][3].

1 储备液制备：
1) 将 500 μg 的 CM-DiI 溶解于 250 μL 的 DMSO 中。
2 工作液制备：
1) 将预热至 37°C 的 7.5 mL Hank's 平衡盐溶液转移至含有 CM-DiI 的 DMSO 溶液中，混合数秒直至染料均匀分布。
3 具体染色步骤
1) 收集过夜的淋巴液，用 Coulter model ZM 细胞计数器测定细胞浓度。取含有 1×10⁹ 个淋巴细胞的体积，在 4°C、300×g 条件下离心 10 min，弃上清，用 HBSS 洗涤沉淀。
2) 将洗涤后的细胞重悬于 2.5 mL HBSS 中，并升温至 37°C。
3) 将上述制备好的染料工作液立即转移至细胞悬液中，通过移液器快速混合，使最终细胞浓度达到1×10⁸ 个淋巴细胞/mL。
4) 细胞在 37°C 孵育 2 min，然后在冰上再孵育 2 min。
5) 将细胞转移至含有 40 mL 4°C HBSS 的 50 mL 离心管中，按照上述条件离心并洗涤。将沉淀重悬于 20 mL 乳酸林格氏液中，将细胞悬液置于冰上，在 15-30 min 内进行注射。
