DiSC3(5) (solution) 

DiSC3(5) (solution) is a fluorescent probe commonly used as a tracer dye to evaluate mitochondrial membrane potential. The excitation/emission wavelength of DiSC3(5) (solution) is up to 622/670 nm. DiSC3(5) can inhibit the respiratory system associated with mitochondrial NAD, and the IC₅₀ value is 8 μM. DiSC3(5) in the presence of Na⁺/K⁺-ATPase inhibitor ouabain 2 can induce membrane hyperpolarization of Ehrlich ascites tumor cells^[1][2][3].

1. 制备 DiSC3(5) 膜染色溶液：
1.1 制备 DMSO 或 EtOH 储备溶液：储备溶液应在 DMSO 或 EtOH 中以 1-5 mM 制备。
注 1)：储备溶液的未使用部分应储存在 -20℃。 避免反复冻/融循环。
1.2 准备工作溶液：将储备溶液稀释到合适的缓冲液中，如无血清培养基， HBSS 或 PBS ，制成 1 至5 uM 的工作溶液。
注2)：对于不同的细胞类型和/或实验条件，应根据经验确定工作溶液的浓度。建议在至少超过十倍范围的浓度下进行测试。
2. 将细胞染成悬浮液：
2.1 在染料工作溶液中悬浮细胞密度为 1×10⁶/ mL。
2.2 在 37°C 孵育 2-20 分钟。 培养时间取决于细胞类型。 首先孵育 20 分钟，然后根据需要进行优化以获得均匀的标记。
2.3 将标记的悬浮管以 1000 至 1500rpm 离心 5 分钟。
2.4 去除上清液，轻轻地将细胞重新悬浮在预热（37°C）的生长培养基中。
2.5 按步骤 2.3 和 2.4 洗涤两次。
3. 染色贴壁细胞：
3.1 在无菌玻璃盖玻片上培养贴壁细胞。
3.2 从生长培养基中取出盖玻片，轻轻地排出多余的培养基。将盖玻片放在湿度箱中。
3.3 将100 μL 染料工作溶液吸移到盖玻片的角落，轻轻搅拌直至所有细胞都被覆盖。
3.4 将盖玻片在 37°C 孵育 2-20 分钟。 培养时间根据细胞类型而变化。开始孵育 20 分钟，然后根据需要进行优化以获得均匀的标记。
3.5 排出染料工作溶液，用生长培养基清洗盖玻片两到三次。对于每个洗涤循环，用预热的生长培养基覆盖细胞，孵育 5-10 分钟，然后排出培养基。
4. 显微镜检测。
5. 流式细胞仪检测：
用 DiS 标记的细胞可使用常规 FL3 流式细胞仪检测通道进行分析。
