ER-Tracker Green (solution) 

ER-Tracker Green (solution) is a derivative of BODIPY series dyes coupled with Glibenclamide (HY-15206), highly selective binding to the endoplasmic reticulum, non-toxic to cells at low concentrations, this type of dye is an environmentally sensitive probe, and formaldehyde treatment can still retain part of the fluorescence, with high fluorescence life, good extinction coefficient and other characteristics. Glibenclamide is an atp-dependent K⁺ channel blocker (Kir6, KATP) and CFTR Cl-channel blocker that binds in the endoplasmic reticulum. ER-Tracker is not suitable for staining cells after fixation^[1].

1. ER-Tracker工作液的制备
1.1 储备液的制备
将100 μg ER-Tracker溶解于128 μL DMSO中，得到1 mM的储备液。
注：推荐贮备液于 -20℃或 -80℃避光保存，避免反复冻融。
1.2 ER-Tracker工作液的配制
将贮备液用无血清稀释细胞培养基或 PBS 以获得 100 nM-1 μM 的 工作液。
注意：ER-Tracker工作液的浓度请根据实际情况调整。
2. 细胞染色
2.1 悬浮细胞 (6孔板)
A。4℃ 1000 g 离心 3-5 分钟，弃上清。PBS洗涤2次，每次5分钟。细胞密度为1×10⁶/mL。
b．加入1 mL工作液，室温孵育5-30分钟。
c． 4℃、400g离心3-4分钟，弃上清。
d．用PBS洗涤2次，每次5分钟。
e．用无血清细胞培养基或 PBS 重悬细胞。荧光显微镜或流式细胞术观察。
2.2 贴壁细胞
a．在无菌盖玻片上培养贴壁细胞。
b.从培养基上取下盖玻片并吸出多余的培养基。
c.加入100 μL工作液，轻摇使之完全覆盖细胞，室温孵育5-30分钟。
d．用培养基洗两次，每次5分钟。荧光显微镜或流式细胞仪观察。
注：若采用流式细胞仪检测，细胞需重悬后再染色。
