FerroOrange is a Fe2+ selective probe with an excitation wavelength of 543 nm and an emission wavelength of 580 nm. FerroOrange emits fluorescence after binding to intracellular Fe2+, thereby achieving specific labeling of Fe2+. FerroOrange is used to detect the content and distribution of Fe2+ in cells[1].

(In Vitro)

指南 (以下是我们推荐的方案，本方案仅提供指南，应根据您的具体需要进行修改)。
1. 溶液配制
1.1 原液配制
溶剂：DMSO
浓度：1 mM。(24 µg FerroOrange 的管中加入 35 µl DMSO，用移液器吹打混匀，配制得到 1 mmol/L FerroOrange 溶液)
储存：分装后，避光保存于 -20°C 或 -80°C。避免反复冻融。
1.2 工作液配制
用 HBSS 或细胞培养基稀释至 1 µM。
注意：工作液应立即配制并使用，并避光保存。

2. 细胞染色
2.1 将细胞接种于 96 孔微孔板中。
2.2 弃去细胞培养基，用 HBSS 或无血清培养基洗涤细胞三次。
2.3 加入 1 µM FerroOrange 工作溶液，37°C 孵育 30 分钟。
2.4 使用荧光显微镜，Ex/Em = 540/585 nm。
