IL-18RAP 蛋白, Rhesus Macaque (HEK293, Fc)

IL-18 is a pleiotropic proinflammatory cytokine that is involved in induction of inflammatory mediators, regulation of the cytotoxic activity of NK cells and T cells, and differentiation and activation of both Th1 and Th2 cells. IL-18 signals through its specific cell surface receptor IL-18R, which comprises two subunits: IL-18Rα and IL-18R beta. IL-18 forms a signalling complex with the IL-18 receptor α (Rα) and β (Rβ) chains at the plasma membrane, which induces multiple inflammatory cytokines^[2]. IL-18R complex recruits the IL-1R- activating kinase and TNF- associated factor 6, which phosphorylates nuclear factor kβ- inducing kinase, with subsequent activation of NF- kβ^[3].IL-18R betaknockdown inhibits IL-18 driven tumor cell metastasis in C57/BL6 mice^[4]. IL-18RAP Protein, Rhesus Macaque (HEK293, Fc) is a recombinant protein with a Fc label that consists of 357 amino acids (M1-R356) and is produced by HEK293 cells.

IL-18Rβ（白细胞介素18受体β，又称IL18RAP）是IL-18R复合物的信号转导链，也是IL-1R家族的成员。IL-18R beta 在 T 细胞、NK 细胞和树突状细胞上表达，但在间充质细胞中不常见表达^[1]。
人 IL-18R beta 蛋白的氨基酸序列与小鼠IL-18R beta蛋白同源性较低。
IL-18R beta是IL-18的受体。IL-18 通过与 IL-18 α 链 (IL-18Rα) 结合形成信号传导复合物。称为 IL-18 受体 β 链 (IL-18Rβ) 的共受体被募集形成高亲和力复合物^[1]。IL-18/IL-18Rα/IL-18Rβ 三元复合物形成并置 IL-18Rα 和 IL-18Rβ 的细胞内 Toll-Interleukin-1 受体结构域。然后，接头分子骨髓分化因子 88 (MyD88) 可能在 TRAM 的帮助下被招募。MyD88 进一步与 IL-1 受体相关激酶 (IRAK) 4 和 IRAK1/2 相互作用，形成称为 Myddosome 的大分子组装体，随后通过 TRAF6 激活 IKK。最后，信号激活 NF-κB 和丝裂原激活蛋白激酶通路7，从而上调各种炎症细胞因子的表达^[2]。
IL-18R beta 与 IL-18 结合，IL-18 受体 α 形成信号复合物，诱导多种炎症细胞因子的表达^[2]。IL-18R beta 敲低可抑制肿瘤细胞转移^[4]。
