Rhodamine-N3 chloride is an azide-rhodamine fluorescent dye that can be used to label biomolecules containing alkyne groups[1][2]. Rhodamine-N3 chloride is a click chemistry reagent, it contains an Azide group and can undergo copper-catalyzed azide-alkyne cycloaddition reaction (CuAAc) with molecules containing Alkyne groups. It can also undergo strain-promoted alkyne-azide cycloaddition (SPAAC) reactions with molecules containing DBCO or BCN groups (Ex/Em = 544/576 nm).

(In Vitro)

原位标记样品处理，基于凝胶的活性蛋白分析[2].
将细胞沉淀在冰上解冻，并在每个样品中加入 200 μL 冷 DPBS 加蛋白酶抑制剂混合物。
裂解细胞，并以 100,000 × g 离心 45 分钟。

对每个样品进行点击化学反应，使用最终浓度为 25 μM Rhodamine-N3 chloride、1 mM TCEP、100 μM TBTA 和 1 mM CuSO4 的溶液，最终体积为 50 μL。
室温孵育 1 小时，后加入 20 μL 4X SDS 上样缓冲液。
然后将样品 (30 μL)上样，在 10% SDS-PAGE 凝胶上进行电泳，并使用荧光扫描仪进行成像。
