Sialidase (α2-3-6-8-9) is a broadly specific sialidase that cuts linear and branched non-reducing terminal sialic acid residues from glycoproteins, glycopeptides, and oligosaccharides. Sialidase (α2-3-6-8-9) can be used for in vitro and in vivo polysaccharide analysis and characterization as well as complete glycoprotein remodeling[1].

本产品由重组大肠杆菌表达。该酶可水解糖蛋白和寡糖上通过 α2-3、α2-6、α2-8 和 α2-9 键连接的唾液酸。α2-3 和 α2-6 键的水解速率高于 α2-8 和 α2-9 键。
储存缓冲液:20 mM Tris,50 mM NaCl,1 mM EDTA,pH 8.0

操作步骤
1. 根据反应体系,加入底物 (2 μg 糖蛋白或 200 nM 寡糖)、1.0 μL 唾液酸酶 (α2-3,6,8,9)、2.5 μL 反应缓冲液 (50 mM CaCl2,500 mM 乙酸钠,pH 5.5),并加水至 25 μL;
2. 混合后,将样品于 37°C 孵育 5-60 分钟。然后于 65°C 加热 10 分钟以终止反应。

注意事项
上述唾液酸酶反应缓冲液的 pH 值为 5.5。如果想用酶法裂解细胞表面的唾液酸,请更换与细胞相容的缓冲液体系。
