T4 DNA 连接酶可催化相邻 DNA 链的 5’-P 末端和 3’-OH 末端以磷酸二酯键结合的反应，需 ATP 作辅酶，不仅可以催化粘性末端之间或平滑末端之间的 DNA 的连接，也可以催化 DNA。
与 NA 之间以及少数 RNA 之间的连接。可应用于在粘性末端或平末端连接双链 DNA 分子，也可应用于修补双链 DNA、RNA 或 DNA RNA 杂交体上的缺口。
反应 buffer：500 mM Tris-HCl，pH 7.5，50 mM MgCl2，25 mM DTT，5 mM ATP
使用说明
1) 按以下组分配制反应体系：
2 µL 反应 buffer；50 ng (0.2-1 pmol) 载体；1-50 ng (0.2-10 pmol) 插入片段；1 µL T4 DNA Ligase；加 ddH2O 补全至 20 µL；
注意：两个片段的连接时，通常载体与插入片段的摩尔比为1:3 (在 1:1-1:10 范围内均可获得理想的实验结果)，提取根据片段大小与浓度，计算其摩尔浓度。
2) 如为粘末端连接反应，22℃ (16-37℃) 连接 30 min，连接产物直接用于转化 (或冻存于 -20℃)。
