EC-17 disodium salt is a folate receptor alpha (FRα) targeting contrast agent with fluorescent properties in the visible light spectrum. The EC-17 maximum excitation and emission wavelengths are 490 nm and 520 nm, respectively[1][2].

1.1 制备储存液[2]
配制 10 μM 的 EC-17 disodium salt 储备液。
注: EC-17 储存液建议分装后于 -20℃ 或 -80℃ 避光保存。
1.2 工作液的配制
用细胞悬液或血液样本稀释储存液，使 EC-17 disodium salt 终浓度为 200 nM。
注:请根据实际情况调整 EC-17 disodium salt 工作液浓度，且现用现配。
2 染色
2.1 体外细胞染色
1) 将 FR+ 的 L1210A 或 KB 细胞以 106 细胞/mL 的浓度悬浮于含 2% 胎牛血清 (FBS) 的 PBS 中。
2) 取 1 mL 细胞悬液，加入 20 μL 的 10 μM EC-17 disodium salt 储备液，使终浓度为 200 nM。
3) 将混合物在 37°C 下孵育 60 min。
4) 用 PBS 洗涤细胞两次，然后以 106 细胞/mL 的浓度重悬，用于后续分析。
2.2 全血样本染色
1) 从雌性裸鼠中抽取血液，用 1000 units/mL Heparin (HY-17567) 以 1:1 比例稀释进行稳定处理。
2) 在 500 μL 血液等分试样中加入 104 个循环肿瘤细胞 (CTCs)，轻轻振荡 30 s 使样本充分混合。
3) 向 0.5 mL 血液样本中加入 10 μL 的 10 μM EC-17 disodium salt 储备液，终浓度为 200 nM。
4) 将血液-细胞悬液在 37°C 下孵育 60 min。
5) 为测试标记特异性，在加入 EC-17 disodium salt 前，先将细胞与 10 μM 游离 Folic acid (HY-16637) 预孵育。
6) 使用流式细胞仪，通过蓝绿色 (488 nm 激光，530 nm 发射滤光片) 和红色 (637 nm 激光，647 nm 发射滤光片) 通道分析标记细胞。

Mice[1]

Mice are injected subcutaneously in the flank with 1.2×106 TC1 cells (C57BL/6 mice), 1.0×106 HeLa cells (NOD.Cg-Prkdcscid  Il2rgtm1Wjl/SzJ mice), or 1.0×106 KB cells (NOD.Cg-Prkdcscid, Il2rgtm1Wjl/SzJ mice). Once tumor volume reached approximately 300 mm3 half of the mice are injected with 0.1 mg/kg of EC-17 and the other half with 0.1 mg/kg of OTL38 via tail vein. Three hours later, the fluorescence of tumors is measured using Flocam[1].
