Resorufin  (Synonyms: 试卤灵; NSC 12097)

Resorufin (NSC 12097) is a highly fluorescent pink dye. Resorufin shows favorable properties such as water solubility, red-shifted absorption/emission signals. Resorufin is used for the detection of ROS/RNS and a second analyte^[1][2].

Resorufin 染色液的配制
1.1 制备储存液
用 DMSO 配制 1000 μM 的 Resorufin。
注：1) Resorufin 储存液建议分装后于 -20°C 避光保存。
1.2 工作液的配制
在 PBS (pH 7.4) 配制成 0.4 μM 的 Resorufin 工作液。
Resorufin 染色
1) 用 CD 预处理 HepG2 细胞 30 min，然后去除培养基。此后，用 200 μL PBS 清洗细胞三次，然后向每个孔中加入 150 μL 新鲜 DMEM 和 15 μL 22 μM 刃天青溶液 (在 DMEM 中稀释)。在 37 °C 黑暗条件下孵育 30 min 后，用平板读数仪测定 Resorufin 含量。
2) 用 CD 预处理 HepG2 细胞 120 min，然后去除培养基。此后，用 200 μL PBS 清洗细胞三次，然后向每个孔中加入 150 μL 新鲜 DMEM 和 15 μL 22 μM 刃天青溶液 (在 DMEM 中稀释)。在 37 °C 黑暗条件下孵育 120 min后，通过平板读数仪测定 Resorufin 含量。
3) 数据分析：采用 Benesi-Hildebrand 方程，其中荧光强度值被吸光度值取代。
