AKR1C3 Antibody (YA2338) is a Rabbit-derived and non-conjugated IgG monoclonal antibody, targeting to AKR1C3.

AKR1C3 是一种胞质醛酮还原酶，能够催化依赖 NADH 和 NADPH 的酮类固醇向羟基类固醇的转化。该酶作为 NAD§(H) 依赖的 3-、17- 和 20-酮类固醇还原酶，在类固醇核和侧链上发挥作用，调控雄激素、雌激素和孕酮的代谢。虽然在体外表现出氧化和还原的双重催化能力，但在体内主要发挥还原酶功能，因其氧化活性会被生理浓度的 NADPH 抑制。AKR1C3 优先作为 17-酮类固醇还原酶，在 AKR1C 酶家族中对 Δ4-雄烯二酮还原为睾酮的催化效率最高。此外，它还能将前列腺素 D2 还原为 11β-前列腺素 F2、孕酮还原为 20α-羟基孕酮、雌酮还原为 17β-雌二醇，并将强效雄激素二氢睾酮 (DHT) 转化为活性较低的 5α-雄烷-3α,17β-二醇 (3α-二醇)。by similar：该酶还表现出对 9-顺式视黄醛的视黄醛还原酶活性。

  Western blot analysis of extracts from A549 (lane 2(20μg), A549 (lane 3(40μg), using AKR1C3 Antibody. Proteins were transferred to a PVDF membrane and blocked with 5% BSA in TBST for 2 hour at room temperature. The primary antibody and Loading control antibody (Beta Actin, HY-P80438, 1/3000) was used in 5% BSA in TBST at 4°C overnight. Goat Anti-Mouse/Rabbit IgG-HRP Secondary Antibody (HY-P8004/HY-P8001, 1/10,000) was used for 1 hour at room temperature.

  Immunocytochemistry analysis of A549 cells labeling AKR1C3 with AKR1C3 antibody (HY-P82593) at 1/50 dilution. Cells were fixed in 4% paraformaldehyde for 15 minutes at room temperature, permeabilized with 0.1% Triton X-100 for 10 minutes at room temperature, then blocked with QuickBlock™ Blocking Buffer for Immunol Staining for 10 min at room temperature. Cells were then incubated with AKR1C3 antibody (HY-P82593) at 1/50 dilution in QuickBlock™ Blocking Buffer for Immunol Staining at 4 ℃. Alexa Fluor® 488-conjugated AffiniPure Goat Anti-Rabbit IgG H&L（HY-P8002, Green) was used as the secondary antibody at 1/1,000 dilution. PBS instead of the primary antibody was used as the secondary antibody only control. The Nuclear counterstain was DAPI (Blue).
