CELF2 Antibody (YA2480) is a Rabbit-derived and non-conjugated IgG monoclonal antibody, targeting to CELF2.

CELF2 是一种 RNA 结合蛋白，参与多种转录后事件的调控，包括 pre-mRNA 的可变剪接（alternative splicing）、mRNA 的翻译和稳定性。它能够介导组织特异性和发育调控的可变剪接，例如在胚胎骨骼肌中特异性激活 TNNT2 外显子 5 的包含，并通过拮抗 PTB 的抑制作用实现这一功能。CELF2 既能激活也能抑制核心调控外显子对，如在 actinin pre-mRNAs 中促进平滑肌（SM）外显子的包含而非肌肉（NM）外显子。此外，它还参与 NMDA 受体 R1 pre-mRNA 外显子 21 的包含和外显子 5 的排除，以及 apoB RNA 编辑活性。在辐射损伤后，CELF2 可增加 COX2 mRNA 的稳定性并抑制其翻译（by similar），并通过调控 COX2 介导的前列腺素 E2（PGE2）表达来调节细胞凋亡程序（by similar）。CELF2 能够结合多种 RNA 序列，如 DMPK 3'-UTR 中的 (CUG)n 三联重复序列、TNNT2 外显子 5 侧翼的肌肉特异性剪接增强子（MSE）内含子位点、富含 UG 的序列（如 UG 重复和 UGUU 基序）、apoB mRNA 中编辑胞苷上游的 AU 富集序列（by similar），以及 COX2 mRNA 3'-UTR 中的 AU 富集序列（by similar）。此外，它可能参与 miRNA 生物合成的调控，例如与 CELF1 共同结合初级 microRNA pri-MIR140 并负调控其成熟过程。

  Western blot analysis of extracts from Jurkat(lane 2(20ug) , THP-1(lane 3(20ug) and HEK293(lane 4(20ug) using CELF2 Antibody (HY-P82735) Rabbit mAb. Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST for 2 hour at room temperature. The primary antibody (1/1000) and Loading control antibody (Beta Actin, HY-P80438, 1/10000) was used in 5% non-fat milk in TBST at 4°C overnight. Goat Anti-Mouse IgG-HRP Secondary Antibody (1/10000) was used for 1 hour at room temperature.

  Immunohistochemical analysis of paraffin-embedded Mouse colon tissue using CELF2 Antibody (YA2480). The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0) for 2 minutes. The tissues were blocked in QuickBlock for 20 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody (HY-P82735, 1/100) in 4℃ overnight. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.
