JAB1 Antibody (YA3159) is a Rabbit-derived and non-conjugated IgG monoclonal antibody, targeting to JAB1.

JAB1 是 COP9 signalosome 复合体 (CSN) 的潜在蛋白酶亚基，该复合体参与多种细胞和发育过程。CSN 复合体通过介导 SCF 型 E3 连接酶复合体 (如 SCF、CSA 或 DDB2) 中 cullin 亚基的 deneddylation，成为 ubiquitin (Ubl) 结合途径的重要调控因子，从而降低其 Ubl 连接酶活性。该复合体还可能与 CK2 和 PKD 激酶结合，参与 p53/TP53、c-jun/JUN、IkappaBalpha/NFKBIA、ITPK1 和 IRF8 的磷酸化。CSN 依赖的 TP53 和 JUN 磷酸化分别促进和保护其被 Ubl 系统降解。在复合体中，JAB1 可能作为催化中心介导 Nedd8 从 cullins 的切割，但其自身无金属蛋白酶活性，需依赖 CSN 复合体的其他亚基。JAB1 直接与大量受 CSN 复合体调控的蛋白相互作用，证实其在复合体中的关键作用，并促进 BRSK2 的蛋白酶体降解。

  Immunocytochemistry analysis of A549 cells labeling JAB1 with JAB1 Antibody (HY-P83414) at 1/50 dilution. Cells were fixed in 4% paraformaldehyde for 15 minutes at room temperature, permeabilized with 0.1% Triton X-100 for 10 minutes at room temperature, then blocked with QuickBlock™ Blocking Buffer for Immunol Staining for 10 min at room temperature. Cells were then incubated with JAB1 Antibody (HY-P83414) at 1/50 dilution in QuickBlock™ Blocking Buffer for Immunol Staining at 4 ℃. Alexa Fluor® 488-conjugated AffiniPure Goat Anti-Rabbit IgG H&L（HY-P8002, Green) was used as the secondary antibody at 1/1,000 dilution. PBS instead of the primary antibody was used as the secondary antibody only control. The Nuclear counterstain was DAPI (Blue).

  Immunohistochemical analysis of paraffin-embedded Mouse testis tissue using JAB1 Antibody (YA3159). The section was pre-treated using heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0) for 8 minutes. The tissues were blocked in QuickBlock for 20 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody (HY-P83414, 1/100) in 4℃ overnight. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.
