LAMP1 Antibody (YA3434) is a Rabbit-derived and non-conjugated IgG monoclonal antibody, targeting to LAMP1.

LAMP1 是一种溶酶体膜糖蛋白，在溶酶体生物发生、溶酶体 pH 调节、自噬 (autophagy) 和胆固醇稳态 (cholesterol homeostasis) 中发挥重要作用。它通过直接抑制质子通道 TMEM175 来调节溶酶体腔 pH，促进溶酶体酸化以实现最佳水解酶活性。LAMP1 还在 NK 细胞毒性中起关键作用，参与细胞毒性颗粒向细胞表面的移动以及穿孔素 (perforin) 向裂解颗粒的运输，并保护 NK 细胞免受自身细胞毒性颗粒内容物引起的脱颗粒相关损伤。此外，LAMP1 向选择素 (selectins) 呈递碳水化合物配体。在微生物感染方面，LAMP1 作为拉沙病毒 (Lassa virus) 糖蛋白的受体，并在酸性较弱的内体中促进病毒与宿主膜的融合。类似功能还包括支持 FURIN 介导的腮腺炎病毒 (mumps virus) 融合蛋白 F 的切割，通过与 FURIN 和未加工的病毒蛋白 F 相互作用实现。

  Western blot analysis of extracts from MDA-MB231 (lane 2(20μg) and NIH3T3 (lane 3(20μg) using LAMP1 (HY-P80206A) Rabbit mAb. Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST for 2 hour at room temperature. The primary antibody (1/1000) and Loading control antibody (Beta Actin, HY-P80438, 1/10000) was used in 5% non-fat milk in TBST at 4°C overnight. Goat Anti-Mouse/Rabbit IgG-HRP Secondary Antibody (1/10000) was used for 1 hour at room temperature

  Immunohistochemical analysis of paraffin-embedded Mouse colon tissue using LAMP1 Antibody (YA3434). The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0) for 8 minutes. The tissues were blocked in QuickBlock for 20 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody (HY-P80206A, 1/10000) in 4℃ overnight. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.

Cells were fixed in 4% paraformaldehyde for 15 minutes at room temperature, permeabilized with 0.1% Triton X-100 for 10 minutes at room temperature, then blocked with QuickBlock™ Blocking Buffer for Immunol Staining for 10 min at room temperature. Cells were then incubated with LAMP1Antibody (HY-P80206A) at 1/50 dilution in QuickBlock™ Blocking Buffer for Immunol Staining at 4 ℃. Alexa Fluor® 488-conjugated AffiniPure Goat Anti-Rabbit IgG H&L（HY-P8002, Green) was used as the secondary antibody at 1/1,000 dilution. PBS instead of the primary antibody was used as the secondary antibody only control. The Nuclear counterstain was DAPI (Blue).
