LEF1 Antibody (YA3164) is a Rabbit-derived and non-conjugated IgG monoclonal antibody, targeting to LEF1.

LEF1 是一种以序列特异性方式结合 DNA 的转录因子。它参与 Wnt 信号通路（By similarity），并在 CTNNB1 和 EP300 存在时激活靶基因的转录（By similarity）。PIAG 可拮抗 LEF1 的 Wnt 依赖性和非依赖性激活（By similarity）。TLE1、TLE2、TLE3 和 TLE4 抑制由 LEF1 和 CTNNB1 介导的反式激活。LEF1 调节 T 细胞受体 α 增强子功能，并通过结合 BLK 的调控位点调节其表达，从而促进 IL17A 表达的 γδ T 细胞成熟和发育（By similarity）。在间充质牙胚形成过程中，LEF1 作为成牙本质细胞分化的正向调节因子发挥作用，但在钟状期被 MSX1 介导的 CTNNB1 信号抑制所抑制（By similarity）。此外，LEF1 可能在毛细胞分化和毛囊形态发生中发挥作用（By similarity）。LEF1 还能转录激活 MYC 和 CCND1 的表达，促进胰腺肿瘤细胞的增殖。某些 LEF1 亚型缺乏 CTNNB1 相互作用域，因此可能作为 Wnt 信号的拮抗剂。LEF1 还能转录激活纤连蛋白启动子，以 CTNNB1 非依赖性方式结合并抑制 E-钙黏蛋白启动子的转录，从而减少胰腺癌细胞的聚集并增加其迁移能力。

  Western blot analysis of extracts from Jurkat (lane 2(20μg) and Jurkat (lane 3(40μg) using LEF1 (HY-P83419) Rabbit mAb. Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST for 2 hour at room temperature. The primary antibody (1/1000) and Loading control antibody (Beta Actin, HY-P80438, 1/10000) was used in 5% non-fat milk in TBST at 4°C overnight. Goat Anti-Mouse/Rabbit IgG-HRP Secondary Antibody (1/10000) was used for 1 hour at room temperature.

  Immunocytochemistry analysis of HT-29 cells labeling LEF1 Antibody (HY-P83419) at 1/50 dilution. Cells were fixed in 4% paraformaldehyde for 15 minutes at room temperature, permeabilized with 0.1% Triton X-100 for 10 minutes at room temperature, then blocked with QuickBlock™ Blocking Buffer for Immunol Staining for 10 min at room temperature. Cells were then incubated with LEF1 Antibody (HY-P83419) at 1/50 dilution in QuickBlock™ Blocking Buffer for Immunol Staining at 4 ℃. Alexa Fluor® 488-conjugated AffiniPure Goat Anti-Rabbit IgG H&L（HY-P8002, Green) was used as the secondary antibody at 1/1,000 dilution. PBS instead of the primary antibody was used as the secondary antibody only control. The Nuclear counterstain was DAPI (Blue).
