Nanog Antibody (YA275) is a Rabbit-derived and non-conjugated IgG monoclonal antibody, targeting to Nanog.

Nanog 是一种转录调节因子，主要参与内细胞团 (inner cell mass) 和胚胎干细胞 (ES cells) 的增殖与自我更新 (self-renewal)。它通过维持 ES 细胞的多能性 (pluripotency) 并抑制其向外胚层 (extraembryonic endoderm) 和滋养层 (trophectoderm) 分化发挥作用。Nanog 能够与 SMAD1 物理结合，干扰共激活因子 (coactivators) 向活性 SMAD 转录复合物的募集，从而阻断骨形态发生蛋白 (bone morphogenetic protein) 诱导的 ES 细胞向中胚层 (mesoderm) 分化。它具有双重功能，可作为转录激活因子或抑制因子，其最优 DNA 结合序列为 5'-TAAT[GT][GT]-3' 或 5'-[CG][GA][CG]C[GC]ATTAN[GC]-3'。Nanog 能够结合 POU5F1/OCT4 启动子，并在分化中的 ES 细胞中通过结合自身启动子 (与 ZNF281/ZFP281 相互作用后) 招募 NuRD 复合体 (NuRD complex) 实现自我抑制 (autorepression)。过表达 Nanog 会促进细胞进入 S 期并增殖。

  Immunohistochemical analysis of paraffin-embedded Mouse testis tissue using Nanog Antibody. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0) for 8 minutes. The tissues were blocked in QuickBlock for 20 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody (HY-P80237, 1/400) in 4℃ overnight. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.
